The NF-E2 DNA binding protein is composed of two subunits, p45 and MafK, and it regulates expression of globin genes in developing erythroid cells through interaction with Maf recognition elements (MAREs). This MAb reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. It is used as tracer agent in CA15. 3 assays. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up-regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion. The dominant epitope of this MAb involves both amino acids as well as sugar moieties. Its epitope is destroyed by desialylation i. e. treatment with Neuraminidase. This antibody recognizes a protein of 35 kDa, which is identified as tartrate-resistant acid phosphatase (TRAcP). It exists as two isoforms (5a and 5b). This MAb reacts with both the isoforms. Recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. This MAb recognizes a protein of 52 kDa, which is identified as TNFR1 (TNFR-associated factor 1). CD30-positive lymphoproliferations of the skin comprise 30% of all primary cutaneous T-cell lymphomas (CTCLs). Besides borderline cases this group includes lymphomatoid papulosis (LyP) and primary cutaneous anaplastic large T-cell lymphoma (cALCL). Although the two entities overlap clinically, histopathologically, immunopathologically and genetically, they differ considerably in their prognosis. In particular, common feature of both cases is histologically the presence of atypical lymphoid CD30-positive T blasts and genetically a clonal T-cell-receptor rearrangement.
